The DNA test was for his dad right? Not the mom...and if it was for his mom, didn't the DNA test results come from a private eye? That guy could have been paid off by Locke's dad and the DNA test to me wouldn't prove anything becuase of that. His dad is a big con and would have pulled out all the stops to make sure that the con worked out...he needed a kidney to stay alive. If they look alike 20 years apart, that doesn't mean too much to me. Al Pacino doesn't look anything at all like he did in the Godfather I lol. But you still didn't answer the question -- was there no doubt at all to Locke that that was his mom?
It does make sense that someone would show up at just the right time for Locke to give his kidney away for his dad.
