Is Charlie Dead?
I do not believe that Charlie is dead. I think Desmond will save him before he dies.
If Charlie died before he got to the station (lightning, drowning, arrow...) he wouldn't of been able to un-block the signal. No one else would of been able to un-block it, because they wouldn't know how to type in the code, since it was made by a musician.
His flashes do focus around Charlie but he has had flashes without Charlie. Desmond said he saw Claire and Aaron get in a helicopter for example.

They can't all be dead because other wise they couldn't kill each other (which they have e.g Libby, Boone, Shannon, Artz, Goodwin, Kelvin, Ethan and Annalucia) Also i wouldn't like to think i had whatched it all this time for it to end them all being dead or a dream or something unreal. it would be a stupid end to a fab show

Tricia Tanaka is Dead
The title of this episode is of course strongly reminiscent of Tom Stoppard's play, Rosencrantz and Guildenstern are Dead, and is similar in that it concerns side characters engaged in activity that is almost entirely inconsequential to the primary story. Conversation on causality and mortality is also prevalent in both works.

Claire. dead or alive
From what you saw, did you think she looked dead or alive. As for me, I think she had an inner peace about her which makes me think she is a spirit, but you never know. Even if she is dead she can always be on the show as a spirit. Look at Christian, he's always around. I have no idea, but I don't think the writers will just keep us hanging and never know what happened to her.

Nikki isn't dead
Nikki isn't dead, although Paulo may be. He was stuck with the spiders longer, and may have more venom in his system.
Nikki's eyes opened right before they covered her head, it's just that no one was looking. So assuming she can survive until the paralysis wears off, she probably will dig herself out. Whether or not she'll bother with Paulo is uncertain.

Now that makes A LOT of sense, at least that it consumed their bodies somehow--the smoke has appeared (possibly) as both Yemi and Christian, and both of their bodies are missing! The question is: Are ALL visions on the island manifestations of the smoke? In that case it doesn't need to consume people to impersonate them, as Walt isn't dead and Dave never existed. Not to mention the horse...
Thinking of all the people who have died on the island, there's really no limit to who can pop up again. Inman's body has never been accounted for...and who can tell if Ana Lucia, Eko, Libby, Shannon, Boone etc. are still in their graves? Was Ethan ever buried? Opens up some interesting possibilities to be sure.
